How to Use the HPLC Mobile Phase Calculator

Proper planning of mobile phase preparation is essential in High-Performance Liquid Chromatography (HPLC) to prevent running dry during a sequence, which can damage the column and pump seals.

Step 1: Enter Method Parameters

Input the Flow Rate (mL/min) and the total Runtime (minutes per injection) as defined in your analytical method (STP).

Step 2: Enter Sequence Details

Enter the total Number of Injections. This should include: System Suitability, Standards, Samples, and Bracketing Standards.

Step 3: Add Safety Overage

It is GMP best practice to prepare excess mobile phase to account for evaporation, priming, and potential re-injections. A standard overage is between 10% to 20%.
